What Rybelsus is

Rybelsus contains semaglutide in tablet form, the first GLP‑1 receptor agonist taken by mouth. It is taken once daily and is used for type 2 diabetes.

Suitability and dosing are decided by a prescriber on the basis of the approved product information.

How it works

Semaglutide, as a daily tablet

Semaglutide is a peptide that would normally be broken down in the stomach. In Rybelsus it is combined with an absorption enhancer (SNAC), which protects it locally and helps it cross the stomach lining.1 Only a small fraction is absorbed, so the way the tablet is taken matters.

On wakingOn an empty stomach, with a sip of water (up to half a glass).
Then waitAt least 30 minutes before eating, drinking or taking other tablets.

Once absorbed, it acts like injected semaglutide on the GLP‑1 receptor. In SOUL, oral semaglutide reduced major cardiovascular events compared with placebo in people with type 2 diabetes and cardiovascular or chronic kidney disease.3

Safety

Side effects and warnings

The most commonly reported side effects are gastrointestinal:

  • Nausea, diarrhoea, vomiting, constipation and abdominal pain, mostly while the dose is being increased
  • Low blood sugar when used with insulin or a sulfonylurea
  • Worsening of diabetic eye disease in people who already have it
  • Very rarely, a condition of the optic nerve (NAION) causing sudden loss of vision has been reported with semaglutide; sudden changes in vision need urgent medical attention.4

Common questions

Do I need a prescription?

Rybelsus is a prescription-only medicine. After your request, the partner pharmacy verifies whether and how supply is possible, for example through an existing prescription or an assessment by an independent doctor where permitted.

How is it different from Ozempic?

Both contain semaglutide. Rybelsus is a tablet taken once a day; Ozempic is an injection given once a week. The doses are not interchangeable.

What are the new and previous formulations?

Rybelsus is available in a newer formulation (1.5, 4 and 9 mg) and in the previous one (3, 7 and 14 mg). The strengths are not interchangeable milligram for milligram; any switch follows the product information.

Why is the dose increased gradually?

Starting low and increasing step by step helps to reduce gastrointestinal side effects. The schedule is set out in the product information and is decided by the prescriber.
